"Hello! This is Lykcan and before we get into the story I want to clarify on this: Yes I do support this ship and no I do not tolerate incest. However many, many people claim to think that these two are distant relatives whether it be direct or indirect. The thing is that I have played all the Adventure games and read the comics (Archie, Sonic the Comic, etc.) and in any of them I have not seen anything correlating to the two being even remotely related.
To start off, in the first Adventure game which was Tikal's only appearance in a main game besides the mobile ones, she basically helps out Sonic and his friends in stopping Chaos from destroying everything. In Knuckles' story he gets flashbacks to which he sees her trying to fend off the echidnas from attacking the Emerald Altar and provoking Chaos. The gameplay in here entails Knuckles finding shards of the Master Emerald to restore it to its former glory. Now in the cutscenes of the two together, there was ABSOLUTELY NO mention of them being relatives whatsoever; the game manual does not even refer them as relatives either. Knuckles was not even aware of who she was and was only surprised that he received a vision from the past.
Most people are unaware of this fact which led me to believe they either have not played the game at all or skipped the cutscenes. I have seen several comments and forums stating that they are indeed related when Sega hasn't even verified or confirmed it. The comics do not even mention them being related as well so the speculation that they are blood-related is entirely false and here's why:
1. Tikal was an only child, born from Pachacamac and her unknown mother. She was not shown to have any siblings, cousins, aunts or uncles. Unless the game specifies relatives that were mentioned but not shown in actual gameplay, Tikal herself only had her father, mother and grandmother with the latter two being deceased, therefore it is IMPOSSIBLE for Knuckles to be her direct descendent. The comics themselves do not mention any of this as well.
2. She never was shown to have children in-game in which she was sealed inside the Master Emerald at a young age. This supports the first statement in which Knuckles is not a descendent of hers as she never became pregnant unless there is some information left out on her normal life, but pertaining to the game events this never happened.
3. In Sonic Chronicles: The Dark Brotherhood, the character Shade mentions Tikal as Knuckles' somewhat distant aunt. This would be an authentic indicator that they are related; however the game was immediately considered non-canon because of the lawsuits that many people know about so it does not apply to the main game continuity. If the claim were to hold and stay canon then of course it would be considered incest, but because it was retconned that means that Knuckles and Tikal are as of now not related at all.
4. If Knuckles and Tikal were to be related, they wouldn't be directly as much due to something called genetic drift. As they are 3,000 years apart their genes would be so different that it would be almost impossible for them to have a near direct bloodline to one another. If for example they were to have kids, they wouldn't have any birth defects or such due to the extreme variation of their genes. Again if they were related I would not have written this but because the games and comics do not directly speculate that they are distant relatives, they are considered to be born from separate families.
I have seen some point out that Tikal is Knuckles' sister or aunt, but that is very farfetched as she was born 3,000 years before the current Sonic timeline and for that to happen, Knuckles would need to have a VERY long lifespan or the egg he was born in (assuming anthropomorphic echidnas can lay eggs if not though this was shown in the comics) had an extremely long incubation period and he hatched centuries later. Some assume this as facts when they are entirely not true as they either have not watched the cutscenes or have not gone that far into looking at their character information. In terms of the timeframe difference, there are several examples of other franchises out there where characters from different timelines hook up together in their canon.
To conclude, no, they are not related in anyway according to the game canon. They come from the same tribe yes but are from separate families as of how it is stated. Until Sega confirms that they are related or an official statement is announced, there is no possibility they are as of the current time. Not everyone has to agree on this ship as they can have others and I do respect that, but for anyone assuming they are related these are the reasons why that is not the case.

Tikal isn't Knuckles' love interest, but are they actually related? Pretty sure the only reason people think that is because of Chronicles.
I don't really think Tikal is related to Knuckles. Ilovejamaa74 did point out some factors, but yeah. Tikal was just a child when she sealed herself into the Master emerald, potentially giving up her corporeal form. She would've had time to produce children.
The only possible way I can see her being related would be if Tikal had any siblings that were not accounted for in Sonic Adventure. Making Tikal as Knuckles great-great-great-etcetera-grand aunt.
That or Pachacamac survived the rising of Angel Island, remarried and had a new heir, but he appeared to be nearing his elderly years at the time, so dunno if Pachacamac would find a new bride, let alone produce an heir.
